TOOS (CAS No. 82692-93-1), with full name N-ethyl-N-(2-hydroxy-3-sulfopropyl)-3-methylaniline sodium salt, is a white or slightly blue crystalline powder.It is an important member of the new Trinder's reagent family and a new highly water-soluble aniline derivative, which is widely used in diagnostic tests and biochemical tests.

It is well known that benzidine derivatives are oxidized by peroxidase and one molecule of hydrogen peroxide. Since the amount of dye produced is proportional to the concentration of hydrogen peroxide, benzidine derivatives are used as diagnostic analysis or enzyme immunoassay as good indicators of hydrogen peroxide.
 
As mentioned earlier, TOOS is an important member of the new Trinder's reagent family. Let's first talk about what the Trinder's reaction is.The Trinder's reaction was first proposed by Trinder in 1969, hence the name Trinder reaction.The principle is that hydrogen peroxide (H2O2) produced by the tested substance through enzymatic action in the presence of 4-aminotelbilin (4-AAP) and peroxidase (POD) enables the chromogenic substance to produce red quinoneimine compounds.Phenol was initially used as a chromogenic agent, and many compounds were later substituted for phenol to significantly improve the sensitivity of the reaction.The comparison and difference between traditional Trinder's reagent and new Trinder's reagent are shown below.The Trinder's reaction forms the basis for enzymatic assays of glucose, cholesterol, triglycerides and uric acid.The chromogenic substance or chromogenic agent mentioned in Trinder's reaction is Trinder's reagent, also known as chromogenic substrate or hydrogen donor.

1  Phenols
The maximum absorption wavelength of the product of the color reaction formed by this kind of Trinder's reagent is 500~520 nm, while the absorption wavelength of bilirubin in serum is 380~530 nm, which will cause interference to the detection results, so it is necessary to add potassium ferricyanide to eliminate interference.


2  Anilines
The maximum absorption wavelength of the product of the color reaction formed by this kind of Trinder's reagent is 550~555 nm, but this kind of color reaction can only be carried out under acidic conditions, and many enzymes will be inactivated under acidic conditions, which greatly limits the breadth of application.
