会社ニュース Application of HEPES in the cultivation of sensitive cell lines (primary cells, stem cells)
Primary cells and stem cells are a difficult category to serve in cell culture. Their tolerance window to the environment is narrow, and a pH drift or osmotic pressure fluctuation may be reflected in the adhesion rate, morphology, or differentiation direction. This also makes the selection of buffer system particularly important in such cultivation.
1.Why are sensitive cells afraid of pH drift
Conventional cell lines have a certain tolerance for short-term pH fluctuations after long-term adaptation. Primary cells come directly from tissues, while stem cells are in an undirected state. The commonality between the two is that metabolism and signaling pathways are more sensitive to environmental changes and have not been "domesticated".
In a bicarbonate containing culture medium, pH depends on the equilibrium with gas-phase CO ₂. When operating outside the incubator, CO ₂ escapes and the pH increases; This window may only cause temporary stress to conventional cells, while it may have irreversible effects on primary cells and stem cells. This is the core reason why HEPES is widely used in such cultivation - its buffering capacity does not depend on gas-phase CO ₂.
2.Application method: Supplement, not replace
In sensitive cell culture, a more secure approach is to use HEPES as a supplement to the bicarbonate system, rather than a replacement:
Maintain the original concentration of sodium bicarbonate in the basic culture medium
Additional 10-25mm HEPES (generic range provided in the manufacturer's technical documentation)
The incubator is dominated by bicarbonate, and when leaving the incubator, it is connected by HEPES
For primary cells and stem cells, it is recommended to measure from the lower edge of the interval (such as 10-15 mM) instead of directly using the upper limit of conventional cell lines.
3.Three variables that must be synchronously controlled
3.1 Osmotic pressure. This is a pit that sensitive cells are prone to step on. The addition of HEPES will increase osmotic pressure, and most mammalian cells can tolerate 260-350 mOsm/kg, while primary and stem cells tend to lean towards the lower part of the range. After adjusting the formula, it must be tested and the dosage of NaCl or NaHCO3 should be correspondingly reduced.
3.2 Lighting. HEPES can generate reactive oxygen species such as hydrogen peroxide under visible light irradiation, especially in the presence of riboflavin in the culture medium. Primary cells and stem cells are already sensitive to oxidative stress, so culture media containing HEPES should be stored away from light and avoid prolonged exposure to strong light during operation. For experiments that require long-term illumination imaging, it can be evaluated to switch to systems such as MOPS.
3.3 Raw material grade. The response of sensitive cells to endotoxins and heavy metal residues is more pronounced. It is recommended to use cell culture grade HEPES for this type of cultivation and focus on the COA indicators of the batch, rather than just the main content.
4 .Suggestions at the operational level
Reduce the exposure time outside the box: Concentrate the steps of liquid change, washing, etc. to shorten the single operation time, which is more effective than simply increasing the buffer concentration
Pre heated medium: reduces the superposition of temperature fluctuations and pH changes
PH calibration at operating temperature: The pKa temperature coefficient of HEPES is approximately -0.014/℃, and it is recommended to calibrate around this temperature when the system operates at 37 ℃
Fixed batch: The experimental cycle for primary and stem cells is long, and the culture medium and buffer should be locked in the same batch as much as possible to reduce variables
Establish your own concentration curve: There are differences in sensitivity between cell types, and it is recommended to conduct a small-scale validation using gradients of 0, 10, 15, 20, and 25 mM, with vitality and morphology as endpoints
5.A premise that needs attention
HEPES is not superior to bicarbonate in all systems. When cultured for a long time in a closed container with poor breathability, the CO ₂ produced by cell metabolism cannot be eliminated, and relying solely on HEPES may lead to a decrease in pH - this is because HEPES does not have the ability to "take away" CO ₂. Therefore, it is more suitable as a supplement to open operations rather than the only buffer source for closed cultivation.
For primary cells and stem cells, the focus of buffer selection is not on "how much to add", but on "controlling variables": taking the lower edge of the concentration range, conducting osmotic compaction testing and review, avoiding light throughout the entire process, and locking in high-grade and stable batches of raw materials. By doing these things properly, pH related fluctuations will be significantly reduced.
