Tris HCl buffer is a commonly used basic reagent in molecular biology and biochemistry experiments. However, many experimenters have some easily overlooked operating habits during use and storage, which may quietly affect the performance of the buffer and interfere with the stability of the experimental results. The following analysis focuses on four typical problems and provides practical improvement suggestions.

There are loopholes in the sealing process, and the risk of volatilization is underestimated
1. Failure to promptly seal the container after use
The water and some components in Tris HCl buffer have a certain degree of volatility. When the container is in an open state, water will continue to evaporate, causing the concentration of the solution to increase and the pH value to shift accordingly. Many experimenters habitually open the container temporarily after use and only remember to seal it after organizing other items, during which time volatilization has already occurred. It is recommended to develop the habit of "sealing as needed" and immediately seal the bottle cap after each liquid extraction.

2. Poor container sealing performance
Some containers on the market have inadequate sealing designs, such as the cap and mouth of cheap plastic bottles not fitting tightly enough, leaving small gaps even after tightening, allowing air to slowly enter and exit. After prolonged storage, the concentration of the buffer solution changes due to slow evaporation. It is recommended to use glass bottles with silicone or rubber sealing rings, or professional sealed plastic containers with guaranteed quality, as these instruments have more reliable sealing performance.
Improper temperature environment control accelerates component loss
1. Long term exposure to high temperature environment
When the buffer solution is placed in a high-temperature area (such as a window sill or near an oven under direct sunlight in summer), the evaporation rate of water increases significantly, and some thermosensitive components may decompose or evaporate rapidly. As short as a few days, there may be a visible decrease in buffering performance. It is recommended to store the buffer solution in a cool, dark reagent cabinet and avoid getting close to heat sources.
2. Sudden temperature changes cause pressure fluctuations
The buffer solution is quickly transferred from a low-temperature environment (such as a 4 ℃ refrigerator) to room temperature, or directly placed in the freezer layer from room temperature. The internal pressure of the container will fluctuate due to sudden temperature changes. This pressure change sometimes opens the bottle cap or causes the sealing ring to briefly fail, thereby accelerating evaporation. When transferring the buffer solution, it can be placed in a room temperature transition area (such as the corner of the experimental table) for 20-30 minutes to allow the temperature to slowly balance before opening for use.
The retrieval operation is not standardized enough and introduces additional interference
1. The tool used for retrieval contains moisture or impurities
When using pipettes, droppers, or measuring cylinders to take liquid, if there are residual water droplets or other solvents inside these tools, the buffer solution will be directly diluted, changing its ionic strength and pH value. Meanwhile, a damp tool surface makes it easier for the buffer solution to adhere and diffuse, increasing the evaporation area. Before each use, it should be confirmed that the tool has been thoroughly dried and there are no visible contaminants. Consider equipping the buffer with a dedicated dry pipette.
During the liquid extraction process, the container remains open for a long time
When using the same bottle of buffer solution multiple times in a row, some operators may open the bottle cap and seal it after all the liquid extraction steps are completed to save time. This approach will continuously expose the buffer to air, and the accumulated volatilization cannot be ignored. Suggest adopting the "packaging" strategy: pre package the commonly used amount of buffer solution into small bottles, only open one small bottle at a time, and keep the large bottle sealed.
Lack of zoning in storage layout, mixed with volatile reagents
Tris HCl buffer should not be stored in the same reagent cabinet as highly volatile chemical reagents such as concentrated hydrochloric acid, concentrated ammonia water, acetic acid, etc. The acidic or alkaline gases generated by the volatilization of these reagents will diffuse into the surrounding environment and undergo slow chemical reactions with the buffer solution, resulting in pH shift or changes in ion strength of the buffer system. In addition, certain gases may also interact with Tris molecules, generating by-products. The correct approach is to set up separate storage areas for volatile acid and alkali reagents, preferably equipped with ventilated reagent cabinets; Tris HCl buffer solution is placed separately in another dry and clean cabinet, with a certain distance between the two.
By paying attention to the four common misconceptions mentioned above and taking corresponding improvement measures, the service life of Tris HCl buffer can be effectively extended, ensuring the repeatability and reliability of experimental results.
