Chemical properties and preparation methods of CHES buffer

The full Chinese name of CHES buffer is 2- (N-cyclohexylamino) ethanesulfonic acid, with the molecular formula C4H10N2O4S and a relative molecular weight of 182.2 g/mol. The appearance is a white crystalline powder that is easily soluble in water at room temperature and has a relatively stable pH value. It exhibits good buffering ability in the pH range of 9.3-9.7, and is therefore widely used in experiments under alkaline conditions.

CHES buffer has the functions of maintaining the stability of the intracellular environment and protecting cells from pH fluctuations. In biology and biochemistry experiments, it can be used for ion buffering, simulating intracellular environment, protein crystallization, electrophoresis experiments, and drug screening experiments.

Characteristics of CHES

1. Chemical stability: CHES has high chemical stability over a wide range of pH values (pH 7.0-9.0), which enables it to effectively maintain the pH value of the solution.

2. Cellular protective effect: CHES has a protective effect on cells, which can prevent them from being affected by external factors such as pH fluctuations and oxidative pressure.

3. Ion buffering ability: CHES has ion buffering ability, which can maintain the ion balance inside and outside the cell, thereby maintaining the normal function of the cell.

4. Biocompatibility: CHES has good compatibility with biological samples and can be used as a buffer for various biological samples.

The configuration method of CHES

The configuration method of CHES is relatively simple, and the specific steps are as follows:

1. Prepare the required reagents: CHES powder, ultrapure water, pH test paper, electronic balance, beaker, magnetic stirrer.

2. Weigh a certain amount of CHES powder and dissolve it in ultrapure water. The specific amount should be determined based on the required buffer concentration and volume. Under normal circumstances, the concentration is 0.1M.

3. Under the action of a magnetic stirrer, slowly add CHES powder to ultrapure water, stirring while adding to ensure complete dissolution of the powder.

4. Measure the pH value of the prepared buffer using a pH test strip. If the pH value is incorrect, it can be adjusted by adding an appropriate amount of acid or alkali. In general, the pH value of CHES buffer should be maintained between 7.0 and 9.0.

5. Transfer the prepared buffer to a clean container, label it with the name, concentration, pH value, and preparation date of the buffer, so that it can be used within its validity period.

Application of CHES

CHES, as an important biological buffer, has a wide range of applications in biological and biochemical experiments. The following introduces some main applications:

1. Maintaining the stability of the intracellular environment: CHES can maintain the stability of the intracellular environment and protect cells from pH fluctuations. In cell culture experiments, the use of this buffer can prolong the survival time of cells and improve their proliferation ability.

2. In vitro simulation of intracellular environment: CHES can be used to simulate the intracellular environment in vitro for various biochemical experiments. For example, when studying enzymatic reactions, using CHES buffer can simulate the pH environment inside cells, thereby obtaining more accurate results.

3. Ion buffering: CHES can be used for ion buffering to maintain the balance of ions inside and outside the cell. In electrophysiological experiments, the use of this buffer can effectively maintain the ion balance of cells, thereby obtaining more accurate electrophysiological data.

4. Biological sample processing: CHES can be used for biological sample processing, such as blood, urine, etc. When processing these biological samples, using this buffer can protect the bioactive substances in the samples from external environmental influences.

5. Other applications: In addition to the above applications, CHES can also be used in protein crystallization, electrophoresis experiments, and drug screening experiments.

It is worth noting that CHES has a certain degree of irritation, and direct contact with the skin or inhalation of its powder should be avoided. When configuring and using, it is necessary to ensure that the operating environment and utensils are clean and sterile.
