Chemiluminescence immunity has replaced enzyme-linked immunoassay to dominate

Chemiluminescence immunoassay is an in vitro diagnostic reagents method developed after enzyme-linked immunization. Commonly used chemiluminescence reagents include luminol, alkaline phosphatase AMPPD, and acridinium esters. Chemiluminescence has high sensitivity, high degree of automation, simple operation, stable reagents and long validity period, but the cost is relatively high. With the development of technology and the reduction of costs, the coexistence of chemiluminescence and enzyme-linked immunoassay has gradually changed.

Chemiluminescence Immunology (CLIA):

It is used for the detection of various antigens, antibodies, hormones, etc., with higher sensitivity than ELISA, strong specificity, wide linear range, stable results, and simplified operation. It is widely used in the detection of clinical specimens. This method has relatively high requirements for equipment, operation, and use environment, and it requires a matching chemiluminescence instrument when used.

Chemiluminescence immunoassay and enzyme-linked immunoassay

Enzyme-linked immunoassay:

It is a highly sensitive immunological experimental technique that combines antigen, antibody-specific reaction and high-efficiency enzyme catalysis on the substrate. In the determination, after adding the enzyme reaction substrate, the substrate is catalyzed into a colored product. The chromogen substrates TOOS, ADPS, MAOS and TMB in ELISA are all commonly used color reagents. Therefore, semi-quantitative or qualitative analysis can be performed according to the shade of color. The detection method has high sensitivity and low difficulty in carrier standardization, but the detection speed is slow, easy to be contaminated, and the steps are relatively complicated.

From the perspective of the international market, the chemiluminescence immunodiagnostic method has basically tested the alternative of the enzyme-linked immunoassay method, and it accounts for nearly 90% of the market share of the immunodiagnostic market. From the statistics of the domestic immunodiagnostic market segment, after 2011, the development of chemiluminescence has accelerated, and the market share has increased from 50% to 73%, occupying an absolute dominant position. In the future, as technology advances and cost control is strengthened, the advantages of chemiluminescence will become more prominent. Taking the international market as a benchmark, there is still a lot of room for development in domestic chemiluminescence methods.

Chemiluminescence CLIA immunoassay has important guiding significance for disease diagnosis, efficacy evaluation, and screening of healthy people. Through accurate quantitative analysis of various trace active substances in human serum, it provides clinicians with an important basis for accurate diagnosis of diseases. As the gold segment of the IVD industry, its market capacity has increased from 2 billion yuan in 2010 to 6.9 billion yuan in 2015, with a compound annual growth rate of more than 25%. With the gradual continuation of chemiluminescence technology changes and substitution trends, and the continuous improvement of overall market demand, it is expected to continue to maintain a market growth of about 25%-30% in the future.

For the analysis of scientific research experiments, if it is qualitative or semi-quantitative analysis, Desheng recommends using enzyme-linked immunosorbent assay, and using TOOS, MAOS for color reagents, etc., if it is quantitative analysis of very small indicators, it is recommended to use chemiluminescence method , Luminol or acridinium ester can be used as luminescent reagent.
