Introduction to Virus transport media solution

Test principle

Viruses are composed of a nucleic acid molecule and protein or only protein. The individual is small and the structure is simple. Because there is no cell structure, the virus cannot replicate itself, but invades its genes into the host cell, using the latter's replication system to replicate new viruses. After the virus sample is collected, in order to maintain the viability of the virus sample and prolong the survival time of the virus in the sample, the sampling swab will be stored and transported in the virus transport media.

virus transport media solution composition and inspection principle

1. The ingredients of the preservation solution are: Hanks liquid foundation, gentamicin, fungal antibiotics, BSA (V), cryoprotectant, biological buffer, amino acid, etc.

2. A combination of multiple antibiotics has anti-bacterial and anti-fungal effects.

3. As a protein stabilizer, bovine serum albumin (BSA) can form a protective film on the protein shell of the virus, making it difficult to decompose and ensuring the integrity of the virus.

4. The neutral environment constructed by Hanks buffer helps increase the survival time and infection stability of the virus.

5. Used for the collection and transportation of clinical influenza, avian influenza (such as H7N9), hand, foot and mouth disease, measles and other virus specimens, as well as mycoplasma, ureaplasma, chlamydia and other specimens

Products and features

This oropharyngeal swab and nasal swab is a commonly used method for obtaining viral samples, which can be used for rapid detection of viral diseases in combination with PCR. However, not every sample collection site can perform PCR detection, so it is necessary to transport the collected virus swab samples. This product is developed for this need and has the following characteristics:

1. Low-temperature non-frozen storage does not damage the outer shell of the virus and is convenient for long-distance transportation.

2. Suitable for all kinds of swab samples, including oral swabs, nasal swabs, throat swabs, etc.

3. It can be used for H1N1 influenza virus and any other viruses that can be sampled with swabs.

4. The viral nucleic acid can be extracted with column virus DNAout or column virus RNAout.

5. The antibiotics in the sampling solution can effectively prevent bacterial and fungal contamination.

6. The sample solution is added with bovine serum albumin, which can protect the virus sample and increase the separation rate

Transport and storage

Transport at low temperature, store at -20℃, valid for one year.

Instructions

Note: The personnel collecting specimens shall take personal protection in accordance with relevant regulations. Pharyngeal and nasal swabs are best collected in the first 3 days of the disease.

1. Dispense this product into a self-prepared 5 mL screw cap plastic tube with a rubber ring in the screw cap (first-level container) in the safety cabinet. Type B product is already in the screw cap centrifuge tube, you can skip this step.

2. Collection of nasal swab: Gently insert the cotton swab into the nasal palate in the nasal passage, and then slowly rotate it out after staying for a while. Swab both nostrils with the same swab. Dip the cotton swab into the screw cap centrifuge tube containing 3 mL of this product obtained in the previous step, discard the tail of the swab, and tighten the screw cap.

3. Collection of pharyngeal swab: wipe the bilateral pharyngeal tonsils and posterior pharyngeal wall with a cotton swab, immerse the cotton swab into the screw cap centrifuge tube containing 3 mL of this product obtained in the first step, discard the swab tail and tighten the screw cover.

4. Write the type of sample, sampling time, number, and patient's name directly on the primary container with an oily marker.

5. Put the sealed specimens in a plastic bag of appropriate size and seal; each bag contains one specimen. At the same time, fill in the relevant information of the specimen on the specimen submission form, and seal it together with the first-level container in a plastic bag.

6. Put the sealed bag containing the specimens into a self-prepared plastic container with a screw cap and a rubber ring (in the secondary container, tighten the cap, and indicate the relevant information on the container. 2 or more sealed specimens of the same patient can be placed In the same secondary container. The secondary container must withstand a pressure of not less than 95 KPa and be lined with water-absorbing and buffering materials. All containers must be printed with biohazard labels.

7. Place the secondary container in a special transport box (or vaccine transport box), put it in the ice tray, fill it with a soft material, and seal it, and transport it by a dedicated person (2 people), not by mail, it is best to use a dedicated vehicle.

8. If the collected specimens cannot be delivered within 24 hours, they should be stored below -70°C as soon as possible. If there is no condition of -70℃, it should be temporarily stored in the refrigerator at 4℃ for a short time, and the specimen should be contacted as soon as possible. Flu virus in

9. It is unstable at -20℃~-40℃, so long-term storage is best performed below -70℃.
