What exactly is ACD solution?

In fact, ACD solution is mainly composed of three components: sodium citrate, citric acid, and glucose, which are suitable for human whole blood testing, blood bank experimental analysis, DNA and paternity testing research, and human leukocyte antigen preservation. Sodium citrate has a high solubility and can act as an anticoagulant, while citric acid has a protective effect against the coking of glucose during disinfection. Glucose is a necessary nutrient in red blood cell metabolism, which can prolong the storage time of red blood cells and prevent hemolysis.

When ACD solution is added to the PRP blood collection tube, it can minimize the error of blood samples, improve the accuracy of detection, not only save time, but also ensure safety and reduce infection.

In addition, when stored at room temperature, the DNA preservation effect with the addition of ACD solution is much better than that of EDTA and heparin, and it will not affect blood samples.

The commonly used ACD solution is a weakly acidic solution with a pH value of 5.0. Usually, 15 grams or 25 grams of ACD solution need to be added for every 0.1 liter of blood. It can be stored for 21 days at a temperature of 4 ℃, and the longest time cannot exceed 28 days.

How to configure ACD solution?

For fresh or frozen blood samples, prepare 0.48g of citric acid, 1.32g of sodium citrate, and 1.47g of glucose, dissolve them in 100ml of double distilled water, and set aside under high-pressure sterilization. Under high-pressure sterilization conditions at 115 degrees Celsius, the pressure can reach 0.075-0.08MPa in 15 minutes. Blood can be stored for several days at 0 ℃ or indefinitely at -70 ℃.
