Application of CAS No 82692-93-1 toos in in vitro diagnostic reagents,Trinder Regent supplier
Toos (CAS No.: 82692-93-1), name: n-ethyl-n - (2-hydroxy-3-sulfopropyl) - 3-methylaniline sodium salt, is a white or slightly blue crystal powder. It is a very important member of the new Trinder's reagent family. It is a new type of highly water-soluble aniline derivative, which is widely used in diagnostic tests and biochemical tests.
It is well known that benzidine derivatives are oxidized by peroxidase and hydrogen peroxide. Because the amount of dye produced is proportional to the concentration of hydrogen peroxide, benzidine derivatives are used as diagnostic analysis or enzyme immunoassay as a good indicator of hydrogen peroxide.
Principle of glucose oxidase peroxidase method
As mentioned earlier, toos is a very important member of the new Trinder's reagent family. Let's talk about the Trinder's reaction first. Trinder's reaction was first proposed by Trinder in 1969.
The principle of this method is that hydrogen peroxide (H2O2) produced by enzymatic reaction of the tested substance can form red quinoline imine compound in the presence of 4-aminotepyrine (4-AAP) and peroxidase (POD). At first, phenol was used as chromogenic agent, and then many compounds were used instead of phenol, which greatly improved the sensitivity of the reaction.
The following is the comparison and difference between the traditional Trinder's reagent and the new Trinder's reagent. Trinder's reaction is the basis of enzymatic determination of glucose, cholesterol, triglyceride and uric acid. In Trinder's reaction, the chromogen or chromogenic agent is Trinder's reagent, also known as color source substrate or hydrogen donor.
|Compare||Traditional Trinder ’s reagents||New Trinder ’s reagent|
|type||Phenol, 4-chlorophenol or sodium 3,5-dichloro-2-hydroxybenzenesulfonate, etc||Sodium aniline sulfonate (ten): ADPS, TOOS, TOPS, ADOS, HDAOS, DAOS, ALPS, MAOS, MADB, TODB|
|Maximum absorption wavelength||500~550nm||＞540nm|
The maximum absorption wavelength of the color reaction product formed by Trinder's reagent is 500 ~ 520nm, while the absorption wavelength of bilirubin in serum is 380 ~ 530nm, which will cause interference to the detection results. Potassium ferricyanide should be added to eliminate the interference
The maximum absorption wavelength of the color reaction products formed by Trinder's reagent is 550 ~ 555nm, but this kind of color reaction can only be carried out under acidic conditions, and many enzymes will be inactivated under acidic conditions, which greatly limits the scope of application.
1. High water solubility;
2. The UV absorption of the color reaction product is more than 540nm;
3. Color reaction requires a wide pH range;
4. High sensitivity of color reaction
It can be seen from the above table that the new Trinder's reagent is superior to the traditional reagent in the actual color reaction. Toos (CAS No.: 82692-93-1) is the most commonly used reagent because of its good performance in absorption wavelength, color reaction intensity and fading.
Compared with the products on the market, TOS produced by Desheng has the characteristics of independent research and development, good crystal shape, pure white crystal color (for Trinder's reaction based on observing color changes, the importance of this is self-evident), and high purity.
