Ethylene diamine tetraacetic acid (EDTA) is an amino polybasic acid. Due to its ability to form complexes with the vast majority of metal ions, it has become a universal strong chelating agent. Although various applications such as extraction, cation exchange, rare element separation, capacity analysis of calcium, magnesium, and barium, softening of hard water, and use as masking agents, metal cleaning agents, detoxifying agents for heavy metal poisoning, and fungicides have been achieved through the synergistic effect of EDTA, the basic research on its structure and synergistic mechanism is still 58 years later.
Tri-potassium EDTA powder
EDTA salt can protect the cellular components of blood, without affecting white blood cell count and size, with minimal impact on red blood cell morphology, and can inhibit platelet aggregation, making it suitable for general hematological testing. Except for the separation and testing of platelets, it is not suitable for coagulation tests and platelet kinetic energy tests, nor for the determination of calcium ions, potassium ions, sodium ions, iron ions, alkaline phosphatase, creatine kinase, and leucine aminopeptidase, as well as PCR tests.
【 Precautions 】
1.The key to preparing whole blood test specimens is sufficient and complete anticoagulation, so after blood collection, it must be reversed 5-8 times as soon as possible to prevent the formation of small blood clots. Especially when the temperature of the blood collection environment is higher than 25 ℃, the mixing of blood and EDTA tripotassium must be timely and sufficient, otherwise it may cause blood coagulation or local coagulation.
2. EDTA salt anticoagulation is irreversible, so the test should be completed within 24 hours after blood collection in EDTA anticoagulant tubes, otherwise it may cause errors in the test results. The storage time of EDTA anticoagulant specimens is: white blood cell count can be stored for 48 hours at 4 ℃, and not more than 24 hours at 23 ℃. Platelet count can be stored for 24 hours at 4 ℃.
3. Anticoagulation requires sufficient EDTA, but excessive EDTA may cause morphological changes in blood cells. Accurately controlling the amount of EDTA tripotassium added is particularly important.
4. EDTA salts can inhibit platelet aggregation, but cannot completely prevent platelet aggregation. A small number of patients may have platelet counts lower than normal in blood routine tests, which may be caused by platelet pseudo aggregation. In this case, sodium citrate anticoagulant can be used for platelet testing verification. This situation is not related to the quality of EDTA anticoagulant, but rather to the patient's blood characteristics and clinical testing procedures.
5. EDTA tripotassium can be used in combination with sodium fluoride and separation gel. When EDTA tripotassium/sodium fluoride is combined, 1.2-2.0mg of tripotassium and 2-4mg of sodium fluoride can be added per milliliter of blood. When EDTA tripotassium is combined with separation gel, the amount of tripotassium added remains unchanged, and the amount of separation gel added to each test tube should be between 0.8-1.2g. When used in conjunction with separation gel, factors such as anticoagulant effect, tube wall silicification, centrifugation conditions, and separation gel quality can all affect the blood separation efficiency. It is recommended to use it in conjunction with the separation gel produced by our company to obtain high-quality plasma samples.
6. After adding to the blood collection tube, it is recommended to use gamma ray irradiation at a dose of 8-25kGy for sterilization. The irradiation dose can be determined based on the initial bacterial count.
The International Committee for Standardization of Hematology (ICSH) recommends EDTA tripotassium salt as the preferred anticoagulant for blood cell counting and classification. Compared with dipotassium EDTA, it has higher solubility and faster anticoagulant speed.
