Preparation Rules Of Tromethamine Tris Buffer

 

 

Tromethamine cas77-86-1, referred to as Tris, commonly known as trimethylol methylamine or trimethylol aminomethane, has a pH range of 7-9 and low ionic strength, which is suitable for most biochemical reactions. Therefore, Tris buffer is widely used in protein electrophoresis, Western blot and nucleic acid agarose gel electrophoresis.

 

Tromethamine Tris has better performance than phosphate buffer, but it is much cheaper than more professional buffer (HEPES, pipes, etc.), so it is a priority to use Tris buffer in biochemical experiments. The dissociation constant pKa of Tris in solution is 8.1 (25 ℃), and the effective buffer range is 7.1-9.1, which can satisfy most biochemical experiments.

Tris solution itself is alkaline, usually combined with acid to form a buffer system, Tris HCl buffer is commonly used. The configuration methods are as follows:

 

Tromethamine (trimethylolmine)

1. Tris reagent powder was dissolved in water, and then mixed with hydrochloric acid solution in different proportions to obtain Tris HCl buffer solution with different pH.

 

2. Directly dissolving Tris HCl reagent powder in water can directly obtain buffer solution with corresponding pH value; Tris HCl bisection reagent is prepared into buffer solution with specific pH value, which can be mixed with HCl solution (or adjusted by Tris solution).

 

When preparing Tris HCl buffer, adjust the pH value with HCl, and then make up the buffer to the required volume. Assuming that there is no overshoot when adjusting the pH value, this method will not change the ionic strength of the buffer.

 

However, after overshoot and adjusting pH with NaOH or Tris HCl and adjusting pH with NaOH, the ionic strength will change because NaOH is a strong electrolyte. Depending on the intended use of Tris buffer, this change may have an impact.

 

In the preparation of Tris buffer solution, it is usually necessary to adjust the pH value, and the electrode compatible with tris should be used when measuring the pH value. When used with tris buffer, the single junction Ag / AgCl electrode may be unstable because silver will gradually precipitate and block the electrode. The use of double junction or calomel reference electrode can ensure accurate pH measurement in Tris buffer without forming precipitation blocking electrode.
