In biological experiments and research, buffer solutions play a crucial role in maintaining the pH stability of the experimental environment, ensuring the accuracy and reliability of experimental results. DIPSO buffer (3- [N, N-di (hydroxyethyl) amino] -2-hydroxypropanesulfonic acid) buffer is a commonly used biological buffer, and its preparation process requires rigorous operating steps and careful attention to details. This article will provide you with a detailed guide on the entire process from DIPSO crystal dissolution to sterilization and storage.

Experimental preparation
1. Material preparation
DIPSO powder
DIPSO crystal powder: Weigh an appropriate amount according to experimental requirements.
Deionized water: used to dissolve DIPSO powder.
Beakers, magnetic stirrers, rotors, measuring cylinders and other experimental equipment.
Sterilization equipment: such as high-pressure steam sterilizers.
2. Safety protection
DIPSO powder has strong hygroscopicity when dissolved in water and is easily decomposed when exposed to light. Therefore, it should be stored strictly sealed, away from light, and kept dry. During the experiment, experimental clothing, gloves, and goggles should be worn to avoid direct contact with the skin and eyes. If accidentally touched, rinse immediately with running water.
DIPSO buffer preparation
1. Dissolve DIPSO
Prepare a quantitative amount of deionized water in a beaker, and the amount of water should be determined based on the volume and concentration of the required buffer solution. Slowly add the weighed DIPSO crystal powder into deionized water to avoid powder splashing. Use a magnetic stirrer to stir until the DIPSO powder is completely dissolved, forming a homogeneous solution.
2. Buffer pH adjustment
Adjust the pH of DIPSO buffer solution to the desired range using acid or alkali solution according to experimental requirements. During the adjustment process, a precision pH meter should be used for monitoring to ensure accurate pH values.
3. Adjust the volume of the solution
Add deionized water until the solution reaches the predetermined volume. Accurately measure the volume of the solution using a graduated cylinder to ensure accurate concentration.
Note that the pH value of DIPSO buffer may be affected by storage temperature and may change during low-temperature freezing, but it does not affect its effectiveness.
Sterilization and storage
1. Sterilization treatment
Pour the prepared DIPSO buffer into the sterilization bottle, being careful not to fill it too full to prevent liquid overflow during the sterilization process.
Use a high-pressure steam sterilizer to sterilize the buffer solution. After sterilization, wait for the buffer solution to cool to room temperature before removing it for use.
2. Storage conditions
The sterilized DIPSO buffer should be stored in an environment of 4-25 ℃ to avoid extreme conditions of high or low temperature.
In order to extend the service life of the buffer solution and maintain its stability, it is recommended to pack it into small bottles and store it below -4 ℃ to avoid repeated freezing and thawing.

Precautions for use
Before using DIPSO buffer, its appearance and pH value should be checked to ensure they meet the experimental requirements. If turbidity, mold or precipitation is found in the buffer solution, it should be immediately discarded. During the experimental process, laboratory safety operating procedures should be followed to ensure personal safety and the accuracy of experimental results.
