HEPES is a kind of Good ’s buffer reagent, which is also called 2-[4-(2Hydroxyethyl)-1-piperazinyl]ethanesulfonic acid. Its pH buffer range is in the range of 6.8 to 8.2. HEPES is difficult to dissolve in any other solvent, but it is easily soluble in water, and there is no precipitation reaction between the same multivalent metal ions. In addition, most of the natural factors will not affect the dissociation of HEPES, such as temperature, concentration, medium salt and ion composition, etc., which shows that the stability of HEPES is very good.

We need to note that the final concentration of HEPES buffer used is 10-50mmol / L, so it does not have any toxicity to the cells. Generally, under normal circumstances, the HEPES concentration can reach the buffer capacity of 20mmol / L. Here are two ways to use HEPES buffer:
The first method of use is that HEPES is directly added to the culture medium that has been prepared according to the required concentration, and then filtered and sterilized. Add 2.38g of HEPES buffer reagent to each thousand milliliters of culture solution. After dissolution, adjust the pH value to 7.2 with 1N sodium hydroxide, and then use it normally after filtration and sterilization. At this time, the concentration of HEPES biological buffer used was 10 mmol / L.
The second method of use usually takes 99 ml of culture solution before use and then adds 1 ml of stock solution. At this time, the application concentration of the solution is still 10 mmol / L. Take 23.8g of HEPES dissolved in 90ml of double-distilled water, adjust the pH to 7.5 to 8.0 with 1N sodium hydroxide, start to dilute to 100ml with water, filter and sterilize, and dispense 2ml vials, the storage condition is 4 ℃ or -20 ℃. This is how to prepare l mol / L (100 x) HEPES stock solution.
