TAPS, fully known as N-tris(hydroxymethyl)methyl-3-aminopropanesulfonic acid, is a zwitterionic buffer, which is widely used in the fields of Biochemistry and molecular biology.It is part of the Tris series of buffers with a pH buffer range of 7.7-9.1 and is soluble in water (25 g/50 ml).As a common buffer system in DNA screening system, TAPS can also be used as a buffer component for RNA samples. TAPS is suitable for the study of electron transfer and phosphorylation in thin-layer preparation of chloroplasts. It protects oxyhemoglobin from oxidizing to methemoglobin during freeze-drying, and can also be used as a background electrolyte for protein microanalysis by capillary zone electrophoresis.
At present, there are few reports on the synthesis process of TAPS, and the synthesis method of TAPS was studied in detail by Chen Guie et al.They reacted with trihydroxymethylaminomethane (Tris) and 1,3-propanesulfonate lactone (1,3-PS) in alcoholic solvents using the following procedures:
1. Weigh equal molar amount of Tris and 1,3-PS, and take appropriate amount of alcohol solvent into 250ml three flasks with condensation reflux. Under stirring condition, heat up and reflux;
2. After a period of reaction, stop the reaction; at room temperature, cool the solution after reaction to room temperature, vacuum suction filtration, TAPS crude filter cake washed with appropriate amount of absolute ethanol for 2-3 times, take out TAPS filter cake, put it into a vacuum drying oven to dry, vacuum suction filtrate recovery for use.

In the above synthesis process, the reaction time, different alcohol solvents and the recycling of reaction solvents have certain effects on the yield and purity of TAPS.The experimental results showed that the highest yield was obtained when the reaction time was controlled at 6 hours, reaching 70.3%, and the yield decreased slightly with the prolongation of the reaction time. For the investigated reaction solvents of ethanol, n-propano
l, isopropanol and n-butanol, the yield and purity of TAPS with n-butanol as the reaction solvent were the highest, 70.3% and 94.36%, respectively. In the reaction process, the recycling of the reaction solvent was conducive to the enhancement of T.Yield and purity of APS.
In summary, TAPS was synthesized using Tris and 1,3-PS as raw materials, reaction time was controlled at 6 hours, n-butanol was used as solvent, and recycled in the reaction process, resulting in the best yield and purity of TAPS.
