Advantages of HEPES buffer in cell culture
HEPES (4-hydroxyethylpiperazine ethanesulfonic acid) is a hydrogen ion buffer with a pH buffer range of 6.8-8.2. In our cell culture process, HEPES buffer is often indispensable. It is a weak acid. In open culture conditions, if HEPES is added, it can prevent the pH from oxidizing and increasing in the medium, thereby maintaining the pH at about 7.0. The final concentration of HEPES buffer is 10-50mmol/L, which has no toxic effect on cells. In general, HEPES with a concentration of 20mmol/L can reach the buffering capacity. Since we are often used in cell culture medium, what are its advantages in cell culture medium?
1. The pH required for most cells is 7.2-7.4, but the optimal pH for cell culture varies with the types of cells being cultured. Fibroblasts prefer a higher pH (7.4-7.7), while the passage of transformed cell lines needs to be biased. Acid pH (7.0-7.4). HEPES is a non-ionic amphoteric buffer, which has a good buffering capacity in the pH range of 7.2-7.4. Its advantage is that it can maintain a relatively constant pH value during open culture or cell observation. Under this culture condition, the lid of the cell culture flask should be tightened to prevent the small amount of carbonate in the culture solution from dispersing into the air .
2. The pH value of the culture medium that uses HCO3 and CO2 buffer pairs is unstable, and it tends to be alkaline after a certain period of storage. If the pH of the culture medium changes too quickly, HEPES buffer can be added to the culture medium to a final concentration of 10--25mM; it can maintain a constant pH for a long time, which is the favorite of cell culturers.
3. Compared with other buffers, the decomposition constant of HEPES does not change much with temperature, which makes HEPES buffer a buffer that can better maintain the structure and function of the enzyme at low temperatures.
However, it is worth noting that when HEPES is exposed to light, H2O2 is produced, which is toxic to cultured cells or other biologically active objects. Therefore, the operation of HEPES as a buffer should be done under dark conditions as much as possible.
