Description
Application scheme of HEPES in mammalian cell culture

In mammalian cell culture, maintaining a stable pH environment is a key factor in determining the growth status of cells and the reliability of experimental results. Although the bicarbonate buffer system is widely used, its strong dependence on the CO ₂ incubator poses a risk of rapid pH drift for cells during lid opening, passage observation, or long-term microscopy imaging. HEPES, as a zwitterionic organic buffer, has become a commonly used solution to address this pain point due to its excellent buffering ability in the physiological pH range.

The Buffer Chemistry Basis of HEPES

The pKa of HEPES (4-hydroxyethylpiperazine ethanesulfonic acid) is approximately 7.31 at 37 ° C and 7.55 at 25 ° C, with an effective buffering range of pH 6.8-8.2, which precisely covers the physiological pH range of most mammalian cells (7.2-7.6). Its buffering mechanism does not rely on the dissolution equilibrium of CO ₂, so it can maintain pH stability even in open culture environments, avoiding alkalization of the culture medium caused by CO ₂ escape.

The molecular structure of HEPES combines sulfonic acid groups and tertiary amine groups, and exhibits an electrically neutral zwitterionic state at physiological pH. It has extremely low permeability to cell membranes, minimal interference with most enzymatic reactions, and almost no chelation of divalent metal ions such as Ca ² ⁺ and Mg ² ⁺. These characteristics make it highly biocompatible in cell culture scenarios.

Application scheme of HEPES in mammalian cell culture

Concentration scheme in conventional cell line culture

The recommended working concentration of HEPES in mammalian cell culture is usually 10-25 mM, and the specific protocol can be flexibly adjusted according to the culture conditions.

Conventional culture supplementation strategy: Adding 10-15 mM HEPES to bicarbonate containing medium can provide an additional pH buffer window for lid opening operations while retaining the buffering capacity of the CO ₂ incubator. This scheme is applicable for daily passaging and observation of conventional adherent or suspended cell lines such as HeLa, HEK293, CHO, etc.

Open operation and imaging scenarios: When cells need to leave the CO ₂ incubator for microscopic observation, live cell imaging, or flow cytometry sorting for a long time, it is recommended to increase the HEPES concentration to 20-25 mM. Studies have shown that RPMI medium buffered with 25 mM HEPES maintains cell viability in long-term culture.

Low serum or serum-free culture: When the serum concentration drops below 1%, the buffering contribution of the serum itself is significantly reduced, and the pH of the culture medium is easily acidified due to lactate accumulation. At this point, 20 mM HEPES can be added to the culture medium as a supplementary buffer. In a low serum Vero cell culture protocol disclosed in a certain patent, the amount of HEPES added is about 2000-3800mg/L, and the culture period can reach 144-200 hours, with a stable pH control in the range of 7.0-7.5.

Preparation method: HEPES powder can be directly added to the culture medium at the target concentration, dissolved and adjusted to pH 7.2-7.4 with NaOH, and then filtered for sterilization. Alternatively, 1 M HEPES sterile storage solution can be pre prepared and added to the culture medium at a ratio of 1:100 before use to achieve a final concentration of 10 mM. HEPES powder itself is resistant to high temperatures, but HEPES solution is not suitable for high-pressure sterilization. It is recommended to use a 0.22 μ m filter membrane for sterilization to avoid the potential impact of high-pressure sterilization on the properties of the buffer solution.

Applicable scenarios and precautions

The advantages of the HEPES solution include long-term operation of cells in biosafety cabinets, CO ₂ - independent culture systems, and sensitive cell line cultures that require precise pH control. But attention should be paid to the following points:

In terms of concentration, HEPES has no significant toxicity to cells within the range of 10-25 mM, but when the concentration exceeds 40 mM, some cell lines may exhibit growth inhibition. In terms of photostability, HEPES may produce trace amounts of hydrogen peroxide under light exposure. For experiments involving fluorescence detection or prolonged light exposure, it is recommended to avoid light and set up a blank control. In addition, the buffering capacity of HEPES comes from its own dissociation equilibrium and does not participate in the metabolic cycle, so it cannot replace the nutritional buffering function of bicarbonate. The optimal strategy is to use the two in combination.
